Glutathione is the least interesting thing NAC does

N-acetylcysteine's reputation begins and ends with antioxidant support for most people. But cysteine, the amino acid NAC delivers, sits at a metabolic crossroads that extends well beyond glutathione synthesis.
Beyond glutathione: cysteine and iron-sulfur cluster assembly
Cysteine is a rate-limiting substrate not only for glutathione, but for iron-sulfur (Fe-S) cluster assembly — the metal-sulfur cofactors embedded in Complex I, Complex II, Complex III, and aconitase. These clusters are built by a dedicated cellular machinery, the ISC (iron-sulfur cluster) pathway, in which a cysteine desulfurase enzyme uses cysteine directly as its sulfur donor. Laboratory studies knocking down components of this pathway show reduced aconitase activity in both mitochondria and cytosol, tracing the effect directly back to impaired Fe-S cluster formation.
Why this reads as broader than "just" an antioxidant
Supporting glutathione protects the electron transport chain and TCA cycle enzymes from oxidative damage after the fact. Supporting Fe-S cluster synthesis is different — it supports the structural cofactors these same enzymes need to function at all. Both trace back to the same cysteine pool, which is why cysteine availability is relevant to more of this system than the "antioxidant" label alone suggests.
Where whey protein fits
Cysteine is one of the few amino acids where dietary form genuinely matters for this pathway. Whey protein delivers cysteine mainly as its dimer, cystine, and several controlled studies using cysteine-rich whey protein isolate have shown measurable increases in plasma or cellular glutathione. For anyone thinking about cysteine status, whey is a well-studied dietary layer underneath supplemental NAC, not a replacement for medical evaluation of any underlying condition.
The takeaway: NAC's connection to Fe-S cluster and aconitase biology is a genuinely under-discussed mechanism worth knowing — best understood as an interesting piece of structural biochemistry, not a stand-alone fix for a struggling metabolic system.
