NAC and Skin Tone: Can a Supplement Really Enhance Your Glow?

How does glutathione affect melanin production?
The mechanism is well established biochemically, and it starts with cysteine's downstream product, glutathione. Glutathione suppresses tyrosinase, the enzyme that initiates melanin production, through two separate routes: direct antioxidant activity that calms the oxidative signaling driving pigment synthesis, and physical binding to the copper atoms that sit at tyrosinase's active site and that the enzyme cannot function without. With tyrosinase activity reduced, the entire pigment-production cascade slows down, and melanogenesis shifts toward a lighter overall output.
What have clinical trials shown about glutathione?
This isn't just theoretical biochemistry — it has been tested directly. A randomized, split-face clinical trial found that topical oxidized glutathione produced a statistically significant, measurable reduction in melanin index, the standard instrument-based metric for skin pigmentation, after 10 weeks of consistent use, with each participant effectively acting as their own control. Separately, several additional randomized, placebo-controlled trials using oral glutathione, at doses ranging between 250 and 500 mg per day, have also reported significant reductions in melanin index compared to placebo groups, suggesting the effect is not limited to topical application.
Do cysteine peptides increase the skin’s resistance to UVB?
There is a second, related benefit worth separating out clearly. A 2024 randomized, placebo-controlled trial specifically testing oral cysteine peptides found a significant increase in participants' Minimal Erythema Dose — the amount of UV-B exposure needed before skin visibly burns. That is a distinct outcome from pigment lightening: it points toward improved UV resilience and reduced sunburn susceptibility, running in parallel with, rather than replacing, any effect on baseline skin tone.
How significant and long-lasting are the effects of supplementation?
The honest caveat, and it is an important one: dermatology systematic reviews still describe the overall evidence base for glutathione as a skin-brightening agent as promising but inconsistent, held back by small sample sizes, short trial durations, and variable methodological quality across studies. The effects reported in well-conducted trials are real and statistically significant, but they tend to be modest in absolute size, and they are not permanent — melanin index measurements generally drift back toward baseline once supplementation stops. This is a legitimate, actively researched corner of dermatology, grounded in a plausible and partially confirmed mechanism, but it is not a proven overnight transformation and shouldn't be marketed as one. Approached with realistic expectations, a reasonable time horizon, and consistent use, it is one of the better-supported "glow" mechanisms currently available — just not a switch you flip once.
