Larazotide Acetate is a first-in-class regulator of intestinal villi tight junction (TJ) being developed as an adjunct to a gluten-free diet. Prevents TJ from opening by promoting TJ folding and actin filament rearrangement, which prevents gluten from entering the intestinal submucosa and triggering an inflammatory response.

Larazotide is an oral peptide derived from zonula occludens that acts as a tight junction regulator and zonulin antagonist. Tight junctions, also known as closure junctions or zonulae occludentes (singular, zonula occludens), are multi-protein junction complexes whose function is to prevent the leakage of solutes and water and to seal between epithelial cells. They also play a key role in maintaining the structure and permeability of endothelial cells. Tight junctions can also serve as leaky pathways, creating selective channels for small cations, anions, or water.

Larazotide binds to gut apical cell receptors and antagonizes zonulin, preventing the opening of epithelial tight junctions caused by gluten/gliadin, glyphosate, pro-inflammatory cytokines, zonulin bacterial antigens such as lipopolysaccharide (LPS) and bacterial dysbiosis.
