Fladrafinil (acetamide, 2-[[bis(4-fluorophenyl)methyl]sulfinyl]-N-hydroxy-) is a bis(p-fluoro) ring-substituted derivative of adrafinil. This means that it has the same effects and mechanisms of action as Adrafinil buthas slightly different biochemical properties. Fladrafinil acts as a selective dopamine reuptake inhibitor (DRI) and has weak affinity for the norepinephrine (NET) and serotonin (SERT) transporters.

The clinical pharmacology of Fladrafinil is similar to that of Adrafinil, but Fladrafinil is approximately 3-4 times more potent than Adrafinil due to the extra fluoride molecule. This causes Fladrafinil to reach peak plasma concentrations faster than Adrafinil. As such, Fladrafinil has a shorter duration of action. Fladrafinil is not currently approved for medical use, so it is classified as a research chemical. It is one of the many cognitive enhancers available on the market today. Fladrafinil acts as a eugeroic drug, i.e. an agent that promotes wakefulness, as a cognitive enhancer. Fladrafinil's mechanism of action has not yet been fully elucidated, but it is believed to work in a similar way to Adrafinil, which is the precursor to Modafinil. Modafinil has been shown to increase hypocretin levels in the brain. Hypocretin is a neuropeptide that regulates wakefulness and arousal, so increasing its levels can help maintain increased alertness, stimulation and wakefulness. Researchers theorize that the effects of Fladrafinil are due to increasing the levels of hypocretin in the brain, as well as dopamine, serotonin and norepinephrine. This, combined with its ability to inhibit the reuptake of these neurotransmitters, has been shown to result in increased levels of alertness, reduced fatigue, and better overall mental performance.
