J-147 is a derivative of curcumin, the active ingredient in turmeric. Unlike curcumin, it can successfully cross the blood-brain barrier.

J-147 works by binding to ATP synthase. Overproduction of ATP is related to the aging process. J-147 can control this, in addition to increasing levels of the neurotransmitters NGF AND BDNF.

Several studies have been conducted with J-147 in mice. A human study was conducted in 2020, but full results have not yet been released.

J-147 AND dementia

Mice with rapidly aging brains were treated with CMS121 AND J147. The researchers found that this reduced cognitive decline, as well as metabolic AND transcriptional markers of aging in the brain. Both compounds preserved mitochondrial homeostasis by regulating acetyl-coenzyme A (acetyl-CoA) metabolism.

Another study in mice showed that by targeting ATP synthase, J147 causes an increase in intracellular calcium, leading to sustained activation of the calcium/calmodulin-dependent protein kinase β (CAMKK2) pathway AMPK / MTOR, a canonical mechanism of longevity.

J-147 I diabetic neuropathy

Diabetic peripheral neuropathy is a condition in which nerve damage occurs as a result of chronically elevated blood glucose levels, as seen in poorly controlled diabetes. J-147 may help treat this complication.  In rats, it was found that J147 can alleviate DPN by negatively regulating AMPK on TRPA1 in vivo AND in vitro. J147 has been suggested as an alternative to the polypharmaceutical approaches required to treat the many pathogenetic mechanisms that contribute to diabetic neuropathy.

J-147 And Depression

J-147 has been studied for its antidepressant effect. It appears to act through cAMP signaling mediated by 5-HT 1A, with improvements observed in rats in just 3 days. In another study in rats, elevated levels of pCREB AND BDNF persisted for 3 hours after a 10 mg/kg dose of J147.

Risks AND side effects of J-147?

J-147 has passed toxicological tests in animal studies as required by the Food and Drug Administration (FDA) AND no adverse effects have been reported in humans when taken in an appropriate daily dose between 10-30mf per day taken with meals.
